ABSTRACT

The application of clustered regularly interspaced short palindromic repeats (CRISPR) and CRISPR-associated proteins (Cas) can be limited due to a lack of compatible protospacer adjacent motif (PAM) sequences in the DNA regions of interest. Recently, SpRY, a variant of Streptococcus pyogenes Cas9 (SpCas9), was reported, which nearly completely fulfils the PAM requirement. Meanwhile, PAMs for SpRY have not been well addressed. In our previous study, we developed the PAM Definition by Observable Sequence Excision (PAM-DOSE) and green fluorescent protein (GFP)‍-reporter systems to study PAMs in human cells. Herein, we endeavored to identify the PAMs of SpRY with these two methods. The results indicated that 5'-NRN-3', 5'-NTA-3', and 5'-NCK-3' could be considered as canonical PAMs. 5'-NCA-3' and 5'-NTK-3' may serve as non-priority PAMs. At the same time, PAM of 5'-NYC-3' is not recommended for human cells. These findings provide further insights into the application of SpRY for human genome editing.

ABSTRACT

Objective:Organizing pneumonia (OP) is a clinical syndrome manifested by granulation tissue plug in respiratory bronchioles, alveolar ducts and alveolar spaces pathologically.With advent of lung biopsy techniques, increasing pathological diagnosis of organizing pneumonia was made, most of which were confirmed secondary organizing pneumonia (SOP) either at time of diagnosis or on follow-up.The secondary etiology is related to the choice of treatment and prognosis.First of all, we need to rule out infection, find evidence of connective tissue disease, or previous medication, radiotherapy and even transplantation.The coexistence of multiple pathological features and simultaneous involvement of pulmonary vessels and pleura suggest possible secondary causes.The pathological diagnosis of organic pneumonia by empty core needle puncture or transbronchial lung biopsy needs to be combined with imaging to avoid omitting the tumor, granuloma and necrosis around organic pneumonia.In general, organizing pneumonia, no matter cryptogenic or secondary, is mostly responsive to corticosteroids compared with other interstitial lung diseases, although relapses are frequent.In summary, real cryptogenic organized pneumonia is actually relatively uncommon and should be diagnosed with caution.Long term follow up is not only crucial to assess the evolution of lung disease but also for looking into the underlying etiologies behind an organizing pneumonia.

ABSTRACT

Objective To introduce a method of automatically identifying critical values from medical image examination reports and prompt the physician to report it,to prevent the omission of the critical value reporting and improve the medical quality.Methods According to the requirement of critical value reporting system,regular expressions were made for each emergency situation of medical image examination,in order to form a critical value feature library.And an algorithm was designed to find critical value and prompt doctors automatically.Results According to this method,the critical value auto recognize software was developed and had been tested in Nanfang Hospital for 6 months.The software ran well.Conclusion Using regular expressions to define a criteria value feature library and design an algorithm of identifying criteria values,can recognize critical values and prompt physician automatically.

ABSTRACT

ECG is a highly practical discipline,not only requires students to have a solid theoretical foundation,but also requires them to have a wealth of clinical practice (graph reading) capacity.This study aims to improve the students' ability of reading ECG,and introduces the game logic and mechanism,to design and implement the educational games APP.The structure of this APP includes learning module,test module,and evaluation and feedback module,which can realize the three functions of independent inquiry,cooperation plus communication,and test challenge.By the development and application of this kind of inquiry medical education game on the mobile terminal,we can provide learners with adaptive,fragmented,and game-based mobile learning environment.

ABSTRACT

This research has been surveyed on street food and snack intake status of elementary school students in the Jeon Buk region and the recognition of mothers has been observed for the objective of providing base materials for appropriate eating habits and nutritional education. As for status on intake of street food, most students (97.3%) have shown to have eaten from the street vendors while mothers assume low frequency in their child's intake of street food. Favorite street food choices had an agreement between mother and student as ddukbokki and dakkochi. Students were more tolerant in sanitation and necessity of street food than the mothers. This research shows that there is a difference in the recognition of street food between the students and their mothers. There a need required for improvement in the system as to provide quality food and to prevent poor quality food being exposed to the students. Furthermore, practical and appropriate education must be conducted at home and at school for better choices of food and a better lifestyle.

ABSTRACT

Objective To develop the system of tumor recognized tool software based on the examination results of multi- tumor protein chip. Methods On the basis of the diagnostic recognized equation, the examination results were comparison by HTA combining VB script and Javascript language program. Results The functions of the software include dates input of tumor protein chip,recognized results, dates interrogation and record printing of 10 kinds of common tumor. Conclusion The software can be diagnosed automatically with the sorts of tumor, which is a scientific, convenient and efficient tool for tumor clinician.
